RESUMO

Red-fluorescing dentine indicates bacterial contamination [Caries Res 2002; 36: 315-319]. We investigated effect of removal of red fluorescent dentine caries on shear bond strength and fracture mode of 4 adhesive approaches. Sixty-five carious teeth and 50 noncarious controls were distributed into 4 groups: Clearfil™ self-etch (CSE), OptiBond™ FL total etch (OTE), Scotchbond™ Universal total etch (STE) and self-etch (SSE). Samples were excited at 405 nm and viewed through 530 nm filter. Carious samples were ground flat exposing strongly red-fluorescing (StrongRF) dentine, on which a composite cylinder was placed, using one of 4 adhesives. After 22 h in water, shear bond strength and fracture mode were analysed. StrongRF was removed; composite cylinders were placed on weakly red-fluorescing (WeakRF) dentine and tested as described above. Finally, red-fluorescing dentine was removed, and composite cylinders were placed on non-fluorescing (NonRF) dentine and tested. Composites were placed at 3 corresponding heights in controls. After 22 h in water, shear bond strength testing and fracture mode analysis were performed. Differences were tested using Mann-Whitney or Wilcoxon tests (p ≤0.05). Median (Q1, Q3) shear bond strength on StrongRF was SSE 14.4 (9.2, 18.2) MPa >CSE 10.2 (6.4, 17.3) MPa >STE 9.1 (6.9, 11.2) MPa >OTE 6.8 (4.0, 10.8) MPa. Shear bond strength increased statistically significantly for all adhesives on WeakRF: SSE 19.8 (13.6, 24.3) MPa >STE 19.5 (12.7, 23.1) MPa >CSE 17.5 (12.0, 22.5) MPa >OTE 15.8 (11.9, 20.9) MPa. Only STE 25.6 (22.4, 29.1) MPa and CSE 22.1 (17.6, 24.6) MPa were significantly different on NonRF compared to WeakRF. For controls tested at corresponding depths, superficial shear bond strength was OTE 18.7 (16.0, 22.1) MPa >STE 18.4 (12.0, 25.9) MPa >CSE 18.1 (12.7, 20.7) MPa >SSE 13.0 (9.6, 17.8) MPa. This was significantly higher compared to StrongRF except for SSE. Central shear bond strength was not significantly different to WeakRF, deep shear bond strength was significantly lower for SSE and CSE but higher for OTE compared to carious. Conclusion: StrongRF dentine should be removed for higher shear bond strength, but WeakRF dentine can often be preserved without compromising adhesive bond strength.

Background: Porcelain fracture or chipping is one of the limitations of all ceramic restorations. This study investigated the shear bond strength (SBS) of composite resins to lithium disilicate ceramics using universal bondings and different methods of surface preparation. Materials and Methods: In this experimental study, 72 specimens of e.max computer-aided design and computer-aided manufacturing (CAD/CAM) ceramic blocks were divided into six groups of 12 according to surface treatment: Group I-Hydrofluoric (HF) acid etching + All-Bond Universal bonding (ABU), Group II-Bur roughening (BR) + HF + ABU, Group III-BR + HF + Bis-Silane (Si) + ABU, Group IV-Sandblasting (SB) + ABU, Group V-SB + HF + ABU, Group VI-SB + HF + Si + ABU. After bonding of composite resin to the prepared ceramic surface and storage of samples in distilled water for 24 h, SBS test was done using the universal testing machine at a crosshead speed of 0.5 mm/min. Data were analyzed using the analysis of variance and Tukey's post hoc test (α = 0.05). Results: The mean values of SBS in six studied groups were 6.65 ± 2.78 MPa, 8.56 ± 2.69 MPa, 8.49 ± 2.14 MPa, 3.13 ± 1.66 MPa, 7.94 ± 2.4 MPa, and 10.04 ± 2.47 MPa, respectively. The mean values of SBS were significantly different (P < 0.001). The highest value of SBS was observed in Group VI and the lowest in Group IV. Conclusion: Ceramic sandblasting followed by HF etching, Bis Si, and ABU resulted in a higher SBS of composite resins to lithium disilicate ceramics.

This narrative review will focus on a number of contemporary considerations relating to the restoration of root filled teeth and future directions for research. Clinicians are now more than ever, aware of the interdependence of the endodontic and restorative aspects of managing root filled teeth, and how these aspects of treatment are fundamental to obtaining the best long-term survival. To obtain the optimal outcomes for patients, clinicians carrying out endodontic treatment should have a vested interest in the restorative phase of the treatment process, as well as an appreciation for the structural and biomechanical effects of endodontic-restorative procedures on restoration and tooth longevity. Furthermore, the currently available research, largely lacks appreciation of occlusal factors in the longevity of root filled teeth, despite surrogate outcomes demonstrating the considerable influence this variable has. Controversies regarding the clinical relevance of minimally invasive endodontic and restorative concepts are largely unanswered with respect to clinical data, and it is therefore, all too easy to dismiss these ideas due to the lack of scientific evidence. However, conceptually, minimally invasive endodontic-restorative philosophies appear to be valid, and therefore, in the pursuit of improved clinical outcomes, it is important that the efficacies of these treatment protocols are determined. Alongside an increased awareness of the preservation of tooth structure, developments in adhesive bonding, ceramic materials and the inevitable integration of digital dentistry, there is also a need to evaluate the efficacy of new treatment philosophies and techniques with well-designed prospective clinical studies.

AIM: The aim of the study was to investigate the influence of cavity cleaning and conditioning on marginal integrity of directly placed post-endodontic composite class-I-restorations in vitro. METHODOLOGY: A total of 168 fully intact teeth without caries or fillings received pre-endodontic composite restorations (class-II) after their extraction. Occlusal endodontic access-cavities were prepared, and root canals were instrumented and filled with gutta-percha and an epoxy resin-based sealer. Prior to post-endodontic class-I-restoration, access cavities were completely contaminated with sealer, cleaned with alcohol and pre-treated as follows: cleaner only (alcohol), glycine-polishing, Al2 O3 sandblasting, carbide bur (immediate as well as delayed restoration). A positive control (not contaminated with sealer and adhesive used) and negative control (cleaner used but no adhesive) were established. Half of the teeth from each group were subjected to thermocycling and mechanical loading (TCML). Marginal integrity of post-endodontic restoration was evaluated in oro-vestibular or mesio-distal sections after AgNO3 dye penetration (DP) by standardized photomacroscopic imaging and expressed in per cent of margin length along all segments and separately for enamel, dentine and composite, respectively. Results were analysed non-parametrically (α = .05). RESULTS: No restorations or teeth fractured or debonded completely. Without TCML, the median DP of all segments was significantly higher for the negative control compared with all other groups in oro-vestibular cutting direction (53%; p = .002) and in mesio-distal cutting direction (51%; p ≤ .041). The other groups without TCML revealed 16%-24% DP (oro-vestibular) and 12%-24% DP (mesio-distal). With TCML, the median DP in oro-vestibular cutting direction for all segments ranged between 48% and 62% for all groups, a significant difference was only observed between glycine-polishing and carbide bur (p = .041). In mesio-distal cutting direction, the median DP in negative control was 69% with TCML and significantly higher compared with all other groups (p = .002). For all other groups, the median DP of all segments ranged between 28% and 40% with TCML without significant differences. Error rates method (k = 7) revealed a significant influence of TCML in general on penetration of all segments in both oro-vestibular and mesio-distal cutting directions. CONCLUSION: Additional access cavity pre-treatment after alcohol cleaning did not improve the marginal integrity of post-endodontic composite restorations. Thorough cleaning of the access cavity with alcohol seems to assure an acceptable marginal integrity to the tooth and restorative composite.

OBJECTIVES: To evaluate the interactions of two phosphate ester monomers [10-methacryloyloxydecyl dihydrogen phosphate (10-MDP) and dipentaerythritol penta-acrylate phosphate (PENTA)] with hydroxyapatite and collagen and understand their influence on dentine bonding. METHODS: Fourier transform infrared spectroscopy, X-ray photoelectron spectroscopy, X-ray diffraction, nuclear magnetic resonance, ultraviolet-visible, and molecular docking were applied for separately evaluating the interactions of two monomers with hydroxyapatite and collagen. Hydrophilicity tests and morphological observation were employed to characterize pretreated dentine. Microtensile bond strength (µTBS) and nanoleakage were investigated to evaluate the bonding performance. Hydroxyproline assay, in situ zymography, and matrix metalloproteinase-9 (MMP-9) activity assay were used to confirm the MMP inhibition. RESULTS: Chemoanalytic characterization confirmed the interactions of 10-MDP and PENTA with hydroxyapatite and collagen. The interactions of PENTA were weaker than 10-MDP. PENTA possessed better dentine tubule sealing after etching than 10-MDP. Dentine treated with PENTA was more hydrophilic than 10-MDP. 10-MDP and PENTA treating significantly increased the initial µTBS than the control group without primer conditioning. µTBS decreased significantly during aging, and the decrease was more severe in the PENTA group than 10-MDP. The 10-MDP and PENTA groups exhibited relatively less fluorescence than the control. The relative inhibition percentages of MMP-9 decreased in the order of 10-MDP-Ca salt, 10-MDP and PENTA. The 10-MDP, PENTA, and 10-MDP-Ca salt groups showed significantly lower hydroxyproline contents than the control. CONCLUSIONS: Although PENTA adsorbed on hydroxyapatite, it did not form a stable calcium salt. The interactions of 10-MDP with hydroxyapatite and collagen are different than those of PENTA. CLINICAL SIGNIFICANCE: The sealing of dentinal tubules by PENTA and the inhibition of MMP by 10-MDP and its calcium salts contribute to improving the dentine bonding durability.

OBJECTIVE: The tooth-resin composite interface is frequently associated with failure because of microbial contamination, hydrolytic and collagenolytic degradation. Thus, designing a dentine bonding system (DBS) with an intrinsically antimicrobial polymerisable monomer is of significance especially if it can be used with self-etching primers enabling resistance to degradation of the interface. METHODS: Experimental adhesives were developed incorporating eugenyl methacrylate (EgMA) at concentrations of 0,10 or 20 wt%, designated as EgMA0, EgMA10 and EgMA20, respectively, for use as a two-step self-etch DBS with the functional monomer bis[2-(methacryloyloxy) ethyl] phosphate (BMEP) in the primer. The curing, thermal and wettability properties of the adhesives were determined, and hybrid layer formation was characterised by confocal laser scanning microscopy, microtensile bond strengths (µTBS) and nanoleakage by back-scattered SEM. In situ zymography was used to assess MMP inhibitory activity of the BMEP-EgMA DBS. RESULTS: EgMA in the adhesives lowered the polymerisation exotherm and resulted in higher Tg, without negatively affecting degree of conversion. Water sorption and solubility were significantly lower with higher concentrations of EgMA in the adhesive. The formation of a distinct hybrid layer was evident from confocal images with the different adhesives, whilst EgMA20 yielded the highest µTBS post water storage challenges and lowest nanoleakage after 6 months. The experimental DBS exhibited minimal to no MMP activity at 3 months. SIGNIFICANCE: The hydrophobic nature of EgMA and high cross-link density exerts considerable benefits in lowering water uptake and polymerisation exotherm. The application of EgMA, adhesives in conjunction with BMEP in a multi-functional self-etching DBS can resist MMP activity, hence, enhance longevity of the dentine-resin composite interface.

OBJECTIVES: The humid oral environment adversely affects the interaction between a functionalised primer and dentine collagen after acid-etching. Robust adhesion of marine mussels to their wet substrates instigates the quest for a strategy that improves the longevity of resin-dentine bonds. In the present study, an etching strategy based on the incorporation of biomimetic dopamine methacrylamide (DMA) as a functionalised primer into phosphoric acid etchant was developed. The mechanism and effect of this DMA-containing acid-etching strategy on bond durability were examined. METHODS: Etchants with different concentrations of DMA (1, 3 or 5 mM) were formulated and tested for their demineralisation efficacy. The interaction between DMA and dentine collagen, the effect of DMA on collagen stability and the collagenase inhibition capacity of the DMA-containing etchants were evaluated. The effectiveness of this new etching strategy on resin-dentine bond durability was investigated. RESULTS: All etchants were capable of demineralising dentine and exposing the collagen matrix. The latter strongly integrated with DMA via covalent bond, hydrogen bond and Van der Waals' forces. These interactions significantly improve collagen stability and inhibited collagenase activity. Application of the etchant containing 5 mM DMA achieved the most durable bonding interface. CONCLUSION: Dopamine methacrylamide interacts with dentine collagen in a humid environment and improves collagen stability. The monomer effectively inactivates collagenase activity. Acid-etching with 5 mM DMA-containing phosphoric acid has the potential to prolong the longevity of bonded dental restorations without compromising clinical operation time. CLINICAL SIGNIFICANCE: The use of 5 mM dopamine methacrylamide-containing phosphoric acid for etching dentine does not require an additional clinical step and has potential to improve the adhesive performance of bonded dental restorations.

A systematic review of the literature was conducted to determine whether immediate dentine sealing (IDS) or delayed dentine sealing (DDS) would lead to a more effective bond between dentine and indirect glass ceramic restorations. Bonding effectiveness was determined by assessing bond strength or force to fracture from the selected studies. This report followed the PRISMA Statement. A total of 10 articles were subsequently identified for inclusion. In vitro studies evaluating bond strength or force to fracture of etchable glass ceramic restorations, following a bonding protocol that incorporates IDS and DDS were eligible to be selected. Analysis has shown IDS creates a more effective bond between dentine and glass ceramic restorations compared with DDS in 9 out of 10 of the studies assessed. Bond effectiveness was judged using three outcomes; microtensile bond strength (µTBS), shear bond strength (SBS) and force to fracture. IDS was shown to increase µTBS and force to fracture, in all seven studies that assessed these outcomes. IDS was also shown to positively affect the SBS in 3 of the 4 studies that assessed SBS. Further clinical trials are now required to understand the significance of IDS in a clinical environment.

OBJECTIVE: To find out if there is a difference in the efficacy of propolis and seventh-generation dentine bonding agent in reducing dentine hypersensitivity. METHODS: The randomised, single-blind study was conducted at the Department of Periodontology, Dow International Dental College, Dow University of Health Sciences, Karachi, from December 2018 to November 2019, and comprised patients with complaint of dentine hypersensitivity who were divided in group A which received 30% ethanolic extract of propolis, and group B which received dentine bonding agent. Recordings of dentine hypersensitivity were obtained at baseline, before and after the application of experimental agents, and on days 7, 15 and 30. The response was measured using the Schiff Cold Air Sensitivity Scale. Data was analysed using SPSS 20. RESULTS: Of the 52 patients, 19 (36.5%) were males and 33 (63.5%) were females. The overall mean age was 29.9 ± 6.5 years. Majority of the subjects were students i.e. 16 (30.8%) and housewives i.e. 11 (21.2%), while drivers, teachers, businessmen etc. constituted of 25 (48%) subjects. Significant reduction of dentine hypersensitivity was observed in both groups (p<0.05). Intergroup comparison showed non-significant differences (p>0.05). CONCLUSIONS: Propolis and dentine bonding agent had significant effect in reducing dentine hypersensitivity. The difference between the two was not significant.

This investigation is vital contribution to the healthcare system utilizing techniques of nanobiotechnology. It interestingly applies chitosan capped CuO nanoparticles in the field of medicine and restorative dentistry. The CuO nanoparticles and CuO-Chitosan nanoparticles are prepared by co-precipitation, and their characterization is performed using X-ray diffraction (XRD), Fourier transform infrared spectroscopy (FTIR), scanning electron microscopy (SEM), transmission electron microscopy (TEM), and energy dispersive X-ray (EDX). The average crystallite size of these nanoparticles has been found to be in the dimensions of <40 nm and <35 nm, respectively. CuO-Chitosan nanoparticles show significant enhancement in in vitro antibacterial, antioxidant, cytotoxic, and antidiabetic activity as compared to CuO nanoparticles. In addition, the successful amalgamation of CuO nanoparticles and CuO-Chitosan nanoparticles into dentine bonding agents results in providing efficient remedy against secondary caries. CuO-Chitosan nanoparticles reinforced dental adhesive discs cause significant upsurge in reduction of Lactobacillus acidophillus and Streptococcus mutans. Also, the augmentation of mechanical properties, water sorption and solubility plus slow and sustained release profile and slight variation of shear bond strength is attained. Taken together, the chemically synthesized CuO nanoparticles and CuO-Chitosan nanoparticles have proven to be promising candidates having enormous potential to be utilized in drug delivery and nanotheranostics.

This vitro study aimed to evaluate the effects of 45S5 bioactive glass (BAG) and Er:YAG laser as desensitization treatments on the microtensile bond strength (MTBS) of fluorosed teeth. The 120 noncarious fluorosis were to obtain superficial dentin, being classified into four groups according to the Thylstrup and Fejerskov Index (TFI). Specimens from each group were randomly divided into five subgroups. After fluorosed teeth hypersensitivity models were established, the following pretreatments were applied on dentine surface: Subgroup 1: deionized water (Control); Subgroup 2: BAG; Subgroup 3: Er:YAG laser; Subgroup 4: BAG + Er:YAG laser, and Subgroup 5: Er:YAG laser + BAG. One sample was randomly selected from each subgroup for scanning electron microscope (SEM). The remaining samples were bonded with composite resin by Adper Single Bond 2 adhesive. Then water bath at 37°C for 24 hr. After 5,000 thermocycling, MTBS was tested and fracture mode was analyzed. The difference of MTBS between BAG group and Control group was found statistically significant (p < .05) in fluorosis. The Er:YAG laser + BAG group showed lowest MTBS values in fluorosis. In conclusion, the pretreatment of BAG might be beneficial to the adhesive of fluorosed teeth. Er:YAG laser desensitization alone or using BAG first and then Er:YAG laser desensitization might not affect the adhesive of fluorosed teeth, while Er:YAG laser desensitization followed by the pretreatment of BAG would be not conducive to the adhesive of fluorosed teeth.

AIM: To compare the shear bond strength of seventh- and eight-generation self-etch dentin bonding agents in primary teeth using universal testing machine. MATERIALS AND METHODS: Forty extracted sound human primary molars were collected and randomly divided into two groups of 20 sample each. Shear bond strength of seventh-generation bonding agent [ADPER Single Bond Universal, (group II)] and eighth-generation bonding agent [Futurabond DC, VOCO Germany, (group I)] were calculated by using Universal Testing Machine (PTC/O83/ME, INSTRON, USA) and expressed in megapascals (MPa). Recorded data were compiled and subjected to statistical analysis using Student's t test. RESULTS: The mean shear bond strength of group I (eighth-generation dentin bonding agent) and group II (seventh-generation dentin bonding agent) ranged from 22.10 to 37.10 MPa and 19.80 to 30.30 MPa, respectively Student's t test showed significantly different and higher (8.7%) shear bond strength in group I when compared to group II (p value = 0.017). CONCLUSION: Shear bond strength of eighth-generation dentin bonding agent (Futurabond DC) was better than seventh-generation dentin bonding agent (Adper Single Bond Universal). CLINICAL SIGNIFICANCE: An effective bond to tooth would reduce marginal microleakage, bacterial penetration, postoperative sensitivity, possibility of pulpal inflammation, and preserve tooth structure by allowing minimal cavity preparation. Current report is paramount contribution via nanotechnology to the existing remedies of health diseases. The lag in application of capped metallic oxide nanoparticles in restorative dentistry exist which is covered by this promising study. The uncapped and chitosan encapsulated ZnO nanoparticles were fabricated by facile co-precipitation method, and characterized using various biophysical strategies including X-ray diffraction (XRD), Fourier transform infrared spectroscopy (FTIR), Scanning electron microscopy (SEM), Transmission electron microscopy (TEM) and Energy dispersive x-ray (EDX). ZnO nanoparticles and ZnO-Citosan nanoparticles were estimated to be <30 nm and <25 nm in size on respective basis. Significant in vitro antibacterial, antioxidant, cytotoxic and antidiabetic activity of ZnO nanoparticles has been elucidated that is enhanced by capping with chitosan polymer. 90% cytotoxicity against brine shrimps, 69.6% antidiabetic activity against α-amylase, and noteworthy antioxidation power by chitosan decorated ZnO nanoparticles has been effectively illustrated. Furthermore, the effective secondary caries remediation approach has been established by an amalgamation of ZnO nanoparticles and ZnO-Chitosan nanoparticles into dentine bonding agents. A remarkable reduction in Streptococcus mutans and Lactobacillus acidophillus strains has been observed, in-specific boosted by chitosan capped ZnO nanoparticles reinforced dental adhesive discs. Additionally, augmented mechanical properties, greater resistance to water sorption and solubility, notably high release profile, and slight variation of shear bond strength values have been obtained. In short, the prepared nanoparticles reported are detected to be auspicious theranostic agents for combating wide array of human pathogens in healthcare system.

OBJECTIVE: To evaluate the feasibility and effect on marginal adaptation in class II composite restoration reinforced with polyethylene fibres in teeth with affected dentine using scanning electron microscopy. METHOD AND MATERIAL: Class II cavities with dimension 1.5 +0 .25 pulpal depth and 4 ± 0.25 mm buccolingual width/.were prepared on proximal surfaces extracted human molars or premolar with affected/sound dentine on gingival margin. The etching priming and restorations were done in prepared samples as per manufacture directions. The samples were divided in 2 groups depending on use of ribbond inserts in composite restorative material. Samples were finished, stored in distilled water and then thermocycled manually and then sectioned longitudinally through the restorations. The marginal adaptation was evaluated using scanning electron microscope (SEM). RESULT: The result showed that there was a definite gap all along the interface between caries affected dentine and the composite material in both the groups. The bigger gap was present in group II compared to group I.

PURPOSE: To evaluate the microtensile bond strength of dentine/self-adhesive resin cement interface after several treatments on a dentine surface. MATERIALS AND METHODS: Twenty-eight human molars were selected and divided into four groups: no treatment (control (C)); 2% chlorhexidine digluconate (CHX); 25% polyacrylic acid (PA); and 23 ppm dispersive solution of silver nanoparticle (SN). Prepolymerised TPH resin composite (Dentsply) blocks were luted on the dentine surface using RelyX U200 self-adhesive resin cement (3M ESPE). Microtensile bond strength was measured (MPa) in a universal testing machine 24 h and 6 months after the bonding process. The fractured specimens were examined in an optical microscope and classified according to the fracture pattern. A representative sample of each group was observed by scanning electronic microscope. Data were submitted to analysis of variance (ANOVA) and Tukey's test to compare the mean among the groups (p <0.05). RESULTS: The highest microtensile bond strength values after 24 h were found for the PA group (13.34 ± 6.36 MPa), with no statistically significant difference for the C group (9.76 ± 3.11 Mpa). After 6 months, the highest microtensile bond strength values were found for the C group (9.09 ± 3.27 Mpa), with statistically significant difference only for the CHX group (2.94 ± 1.66 MPa). There was statistically significant difference only for the PA group when comparing the periods studied. Regardless of the surface treatment applied, there were more adhesive failures in both periods of time. CONCLUSION: Dentinal pretreatment with PA, as well as use of SN before the bonding procedure of self-adhesive resin cement to dentine, may be alternative bonding protocols.

The insufficient radiopacity of dental adhesives applied under composite restorations makes the radiographic diagnosis of recurrent caries challenging. Consequently, the misdiagnosis may lead to unnecessary replacement of restorations. The aims of this study were to formulate experimental dental adhesives containing cerium dioxide (CeO2) and investigate the effects of different loadings of CeO2 on their radiopacity and degree of conversion for the first time. CeO2 was characterized by X-ray diffraction analysis, Fourier transforms infrared spectroscopy, and laser diffraction for particle size analysis. Experimental dental adhesives were formulated with CeO2 as the inorganic filler with loadings ranging from 0.36 to 5.76 vol.%. The unfilled adhesive was used as a control. The studied adhesives were evaluated for dispersion of CeO2 in the polymerized samples, degree of conversion, and radiopacity. CeO2 presented a monoclinic crystalline phase, peaks related to Ce-O bonding, and an average particle size of around 16 µm. CeO2 was dispersed in the adhesive, and the addition of these particles increased the adhesives' radiopacity (p < 0.05). There was a significant decrease in the degree of conversion with CeO2 loadings higher than 1.44 vol.%. However, all materials showed a similar degree of conversion in comparison to commercially available adhesives. CeO2 particles were investigated for the first time as a promising compound to improve the radiopacity of the dental adhesives.

Abstract The most common main materials for dentin bonding for composite resin restoration is 2-hydroxyethyl methacrylate (HEMA). HEMA has beneficial physical and chemical properties, and stable, yet toxic. The addition of ethanol or water, may reduce the toxic effect of HEMA. Ethanol solvent has lower H-bonding capacity compared to water solvent, so it can bind less free radicals from the residual monomer. This study aimed to analyze apoptosis due to dentine bonding application with ethanol and water solvent. Fibroblast culture cells were obtained from extracted third molar, by means of tripsinasion method. The cells were divided into 4 groups as reached confluent: cell culture without treatment as control, cell culture with scaffold chitosan, cell culture with scaffold and polymerized dentin bonding with ethanol or water solvent. Apoptosis observation was conducted using immunohistochemistry method with ethidium bromide acridin orange staining, under fluorescent microscope with 40´ magnification. There was a significant difference among groups (p=0.0001), yet no differences found between different solvent. Apoptosis rate in fibroblast cells culture exposed to HEMA bonding with ethanol solvent was 67%, while the cells exposed to HEMA bonding with water solvent was 44%. The effect of dentin bonding with ethanol solvent and water solvent towards apoptosis rate of pulp fibroblast cells is not different.


Resumo Os principais materiais para adesão dentinária em restaurações de resina composta são o 2-hidroxietil metacrilato (HEMA). O HEMA possui propriedades físicas e químicas benéficas e estáveis, ainda que tóxicas. A adição de etanol ou água pode reduzir o efeito tóxico do HEMA. O solvente etanol possui uma menor capacidade de ligação H em comparação com o solvente água, de modo que pode ligar menos radicais livres do monômero residual. Este trabalho teve como objetivo analisar a apoptose pela aplicação de adesivos dentinários com solventes etanol e água. Células de cultura de fibroblastos foram obtidas a partir do terceiro molar extraído, por meio do método de tripsinaion. As células foram divididas em 4 grupos como confluentes: cultura celular sem tratamento como controle, cultura celular com arcabouço de quitosana, cultura celular com arcabouço e adesivo dentinário polimerizado com solvente etanol ou água. A observação da apoptose foi realizada utilizando o método imunohistoquímico com coloração com brometo de etídio e acridina laranja, sob microscópio de fluorescência com aumento de 40´. Houve uma diferença significativa entre os grupos (p = 0,0001), mas não houve diferenças entre os solventes. A taxa de apoptose em cultura de células de fibroblastos expostos à adesão baseada em HEMA com solvente etanol foi de 67%, enquanto as células expostas à adesão baseada em HEMA com solvente de água foi de 44%. O efeito da adesão dentinária com solvente etanol e solvente água sobre a taxa de apoptose de células de fibroblastos de polpa não é diferente.

This study aimed to evaluate the effects of two calcium-containing desensitizing pastes on the bonding stability of an etch-and-rinse (E&R) adhesive to dentine. After dentine hypersensitivity model established, dentine surfaces were assigned one of the following pretreatment: Group 1, no desensitizer; Group 2, CPP-ACP; and Group 3, Novamin. Specimens were then bonded with an E&R adhesive. Beams from each tooth were randomly divided into three subgroups and then subjected to microtensile bond strength (MTBS) test after 24 h; 12 months of water storage; or 15 runs of pH cycling. Failure modes, nanoleakage, and tubule-occluding effectiveness were analyzed. Results showed that CPP-ACP- or Novamin-pretreated specimens mainly preserved the bonding strength after 12 months of water storage, while effective tubule occlusion could be observed. The results suggested that the calcium-containing desensitizers were compatible pretreatment for bonding with E&R adhesives to obtain reliable long-term bonding strength and prevention of post-operative sensitivity.

OBJECTIVES: To evaluate the effect of intrinsic water permeation on the microtensile bond strengths of different adhesive systems to dentine and the quality of resin-dentine interfaces. METHODS: Ninety-six non-carious human third molars were divided into 4 groups: Clearfil S3 Bond Plus (CSBP; Kuraray); Clearfil S3 Bond (C3S; Kuraray); iBond Self-Etch (IB; Heraeus-Kulzer) and Prime&Bond NT (PB, control etch-and-rinse adhesive, Dentply-Sirona). For each adhesive, specimens from one subgroup (N=10) were bonded using zero pulpal pressure, while specimens from the other subgroup (N=10) were bonded using 15cm water pressure (PP). Each bonded tooth was sectioned into 1×1mm sticks and stressed to failure. Data were analysed using two-way ANOVA and Holm-Sidak pairwise comparisons to examine the effects of "adhesive", "pulpal pressure" and their interaction on bond strength (α=0.05). Representative fractured sticks were examined by SEM. The remaining tooth slabs in each subgroup were used for TEM and CLSM. RESULTS: Microtensile bond strengths (mean±SD; in MPa) were: 33.4±6.9 (CSBP), 33.2±4.7 (CSBP-PP), 35.0±8.6 (C3S), 25.5±7.3 (C3S-PP), 18.4±4.0 (IB), 16.5±6.9 (IB-PP), 28.2±5.5 (PB), 20.5±7.2 (PB-PP). "Adhesive-type" (P<0.001), "pulpal-pressure" (P<0.001) and their interactions (P<0.001) significantly affected bond strength results. No difference between no-PP and PP subgroups was found for CSBP and IB (P>0.05). Water droplets were identified along the resin-dentine interface for IB, IB-PP and C3S-PP. CONCLUSION: IB exhibits water sensitivity when bonding is performed with/without pulpal pressure. C3S exhibits water sensitivity when bonding is performed with pulpal pressure. CSBP does not exhibit water sensitivity when bonding is performed with/without pulpal pressure. CLINICAL SIGNIFICANCE: Intrinsic water permeation during bonding procedures significantly affects bond strength results and the resin-dentine interface of contemporary single-bottle self-etch dentine adhesive systems.

Abstract The aim of this study was to formulate and evaluate an experimental adhesive resin with the addition of 1,3,5-triacryloylhexahydro-1,3,5-triazine at different concentrations. Experimental adhesive resins were obtained by mixing 50% wt bisphenol A glycol dimethacrylate (BisGMA), 25% wt triethylene glycol dimethacrylate (TEGDMA), 25% wt 2-hydroxyethyl methacrylate (HEMA) and photoinitiator system. The triazine compound was added in 1, 2.5 and 5% wt to a base adhesive resin and one group remained with no triazine as control group. The experimental adhesive resins were analyzed for antibacterial activity (n=3), degree of conversion (n=3) and softening in solvent (n=3). Data distribution was evaluated by Kolmogorov-Smirnov test, paired t test, one-way ANOVA and Tukey’s with a 0.05 level of significance. All groups with added triazine compound showed antibacterial activity against Streptococcus mutans (p<0.05). All groups achieved more than 70% degree of conversion, but there was no difference in this chemical property (p>0.05). The initial Knoop hardness was higher in 2.5 and 5% wt groups (p<0.05) and both groups present lower percentage variation of Knoop hardness after solvent degradation. The present study formulated an antibacterial adhesive resin with a non-releasing agent able to copolymerize with the comonomeric blend, improving the restorative material’s properties.


Resumo O objetivo desse estudo foi desenvolver e avaliar a adição de 1,3,5-triacryloylhexahydro-1,3,5-triazine a uma resina adesiva experimental em diferentes concentrações. Resinas adesivas experimentais foram obtidas a partir da mistura de 50% em peso de bisphenol A glycol dimethacrylate (BisGMA), 25% em peso de triethylene glycol dimethacrylate (TEGDMA), 25% em peso de hidroxietil metacrilato (HEMA) e sistema fotoiniciador. O composto de triazina foi adicionado em proporções de 1; 2,5 e 5% em peso a resina adesiva base e um grupo permaneceu sem a adição do composto de triazina como grupo controle. As resinas adesivas experimentais foram analisadas por atividade antibacteriana (n=3), grau de conversão (n=3) e degradação em solvente (n=3). A distribuição dos dados foi avaliada por teste de Kolmogorov-Smirnov, teste t pareado, ANOVA de uma via e Tukey, considerando nível de significância de 5%. Todos os grupos com adição de composto de triazina demonstraram atividade antibacteriana contra Streptococcus mutans (p<0,05). Todos os grupos atingiram mais de 70% de grau de conversão, mas não houve diferença estatística para essa propriedade química (p>0,05). A dureza Knoop inicial foi maior para os grupos com 2,5 e 5% de triazina (p<0,05) e ambos os grupos apresentaram menor variação percentual de dureza Knoop após degradação em solvente. No presente estudo, foi produzida uma resina adesiva antibacteriana com agente sem liberação para o meio, capaz de copolimerizar com a blenda comonomérica, melhorando as propriedades do material restaurador.
